Controlled Release of Transforming Growth Factor-beta1 Enhances Chondrogenesis Ofcollagen/chitosan/gag Scaffold

چکیده
INTRODUCTION: Transforming growth factorβ1(TGF) has been reported to influence the proliferation of chondrocytes and also stimulate proteoglycan synthesis. However, this chondrocyte’s response to TGF can be achieved through prolonged exposure to TGF. Therefore, a focal application of controlled releasing system remains attractive for enhancing chondrogenesis of tissue engineered constructs. Porous collagen based scaffold has a great potential in cartilage tissue engineering. Chondroitin sulfate (CS), a component of glycosaminoglycans (GAG) present in articular cartilage, and chitosan similar with various GAG are also used as biomaterials available to stimulate chondrogenesis. In this study, in order to develop a novel tissue engineered cartilage, TGF loaded chitosan microspheres were incorporated into the porous collagen/chitosan/CS scaffolds. In addition, we evaluated whether TGF loaded chitosan microspheres based delivery systems modulate controlled release of TGF in a manner that favorably influence chondrocyte’s chondrogenesis.

Human intervertebral disc (IVD) degeneration often initiated from the human nucleus pulposus (hNP) with aging leading to IVD destruction and extracellular matrix (ECM) depletion. Previously, we have successfully employed transforming growth factor-beta1 (TGF-beta1) to promote chondrogenesis of mesenchymal progenitor cells (MPCs) and immortalized human mesenchymal stem cells. In this study, we e...

Regeneration deficiency is one of the main obstacles limiting the effectiveness of tissue-engineered scaffolds. To develop scaffolds that are capable of accelerating regeneration, we created a heparin/chitosan nanoparticle-immobilized decellularized bovine jugular vein scaffold to increase the loading capacity and allow for controlled release of vascular endothelial growth factor (VEGF). The va...

To examine the effect of transforming growth factor (TGF)-beta1 on the regulation of cartilage synthesis and other articular pathologies, we used adenovirus-mediated intra-articular gene transfer of TGF-beta1 to both naïve and arthritic rabbit knee joints. Increasing doses of adenoviral vector expressing TGF-beta1 were injected into normal and antigen-induced arthritis rabbit knee joints throug...
